What Is GHK-Cu? The Copper Peptide, Topical and Otherwise

GHK-Cu is unusual among the compounds in this space because it has a substantial presence in two very different worlds: it is a well-established cosmetic skincare ingredient sold over the counter, and it is also prepared as a compounded injectable. The evidence supporting those two uses is not equally strong, and conflating them is the most common mistake in how this compound gets discussed.

The short answer

GHK-Cu is a naturally occurring tripeptide — glycyl-L-histidyl-L-lysine — bound to a copper ion. It is present in human plasma, saliva, and urine, and plasma concentration declines substantially with age. Research has examined roles in wound healing, collagen and elastin synthesis, and antioxidant activity. The strongest evidence relates to topical application; injectable preparations are compounded and less extensively studied.

A peptide your body already carries

GHK is a three-amino-acid sequence — glycine, histidine, lysine — with a strong natural affinity for copper. In the body it exists predominantly bound to copper as GHK-Cu, and it is found in plasma, saliva, and urine.

The frequently cited observation is the age-related decline: plasma GHK concentration is roughly 200 nanograms per milliliter around age twenty and falls to approximately 80 by age sixty. That decline correlates with the period during which skin regenerative capacity and wound-healing speed also decline, which is the origin of the interest in the compound.

Correlation is doing real work in that sentence, and it should be read as the reason the research was pursued rather than as the conclusion of it.

What the topical research supports

The best-developed evidence for GHK-Cu concerns topical application to skin. Studies have examined effects on collagen and elastin synthesis, glycosaminoglycan production, skin firmness and elasticity measures, and wound repair. This is why copper peptides appear as a named active ingredient across a wide range of serums and creams — the cosmetic use rests on a real literature.

Because topical GHK-Cu is a cosmetic ingredient rather than a drug, it does not require a prescription and its claims are cosmetic claims. Injectable preparations are a different question

Injectable GHK-Cu is a compounded preparation, prescribed individually and produced by a compounding pharmacy. It has not been through FDA pre-market review in that form, and the human clinical literature supporting systemic administration is considerably thinner than the topical dermatologic literature.

This is worth stating plainly because the marketing frequently borrows credibility across the boundary — citing topical skin studies in support of an injectable protocol. Those are different exposures, different tissue concentrations, and different evidence bases. A physician evaluating whether an injectable preparation is appropriate is making a judgment that the topical research does not settle.

Where it fits alongside aesthetic care
